Assay of the Multiple Energy-Producing Pathways of Mammalian Cells
OmniLog provides important advantages over end-point reads on a microplate reader in kinetic reading and throughput.
Although end-point reads (when read at the proper time) can give partial quantitative information, they are usually just one data point, a non-averaged value with high statistical variance.
The best information from the PMM assays is obtained by measuring initial rates of energy production, based on multiple reads. Using Biolog's proprietary tetrazolium dyes, the kinetics of dye reduction and purple color formation are initially linear with time. The OmniLog and OmniLog Software allow you to truncate the assays to their linear phase and accurately determine these initial metabolic rates for each well of each plate. Furthermore, dye reduction rates are independent of initial well color density, which may vary. Using OmniLog, no preliminary experiments need to be performed to determine the optimal time window for data collection because the kinetic record contains the entire time course. The kinetic read capability enables the relative rates of energy flux generation (or alterations) to be assigned to each substrate or metabolic effector.
Throughput is a second major advantage. OmniLog has a capacity to assay 16 plates at 5 minute read intervals, or 50 plates at 15 minute read intervals.The large 50 plate capacity of the OmniLog enables multiple experiments (incorporating different media formulations, cell types, or chemical agents) to be run simultaneously.
The software allows differences in substrate metabolism and rates of metabolism to be quickly assessed and displayed. Each pre-loaded substrate has a KEGG database identifier number. The software allows substrates of interest to be quickly matched to their respective pathway in KEGG. Data collected over many months of experiments are stored in standard formats and can be readily retrieved and compared.
